The city of DuQuoin inherited its name from an old Kaskasian Indian named Chief Jean Baptist Du Coigne, Chief of the Kaskasian Indian Tribe. Chief Du Coigne died in 1811 and is buried in Kaskaskia.

DuQuoin was plotted in 1853, and being located next to the mainline of the newly built Illinois Central Railroad, it was an overnight success.

There were numerous settlers in the surrounding area, but the first settlers where DuQuoin stands today, were Colonel John and Abner Pyle in 1819. Daniel Dry was the first person to open a store in 1830.

B.G. Root and Louis Wells surveyed the section of land running through DuQuoin for the railroads. In 1850, Isaac S. Metcalf, a civil engineer, built the railroad, finishing it in 1855.

In 1853, Isaac S. Metcalf and Chester A. Keyes laid out a new town by the railroad and called it DuQuoin. It was a 40 acre tract platted by William Richart, a surveyor from Jackson County. DuQuoin was officially dedicated on September 20, 1853.

Du Quoin participates in an Enterprise Zone configuration with Perry County, the City of Pinckneyville and the Village of St. Johns.
